Yes, come 2012, there will be a new Spider-Man film.
It will be a reboot and it will be directed by Marc Webb. Andrew Garfield will be playing a younger Peter Parker. Many people believe that this will be based on the Ultimate Spider-Man series written by Brian Michael Bendis. Sony still has the film rights to Spider-Man, so he will not be involved with Marvel's Avengers or their universe.
If it were up to me, I'd skip the origin (as we all know who Spider-Man is by now) and go straight into a story involving the Vulture and Mysterio as the main villains. Gwen Stacy should be Parker's love interest. Mary Jane can be introduced, but she's better saved for a sequel.
I've thought out a whole chain of these movies just for kicks, but I know they'd never go with this:
Spider-Man: Vs. Vulture & Mysterio, Gwen Stacy, Osborns introduced
Spider-Man 2: Vs. Green Goblin, Daily Bugel and Otto Octavius introduced, Death of Gwen Stacy
Spider-Man 3: Vs. Doctor Octopus & Electro, MJ is introduced, Peter graduates from high school, Harry Osborn tries to kill himself.
Spider-Man 4: Vs. Kraven & Lizard, Peter goes to college and begins to have a relationship with MJ, Harry is recovering slowly
Spider-Man 5: Vs. Venom, Sandman (as a smalltime villain) and Black Cat introduced, Peter proposes to MJ, sets up for a Venom spin-off
Spider-Man 6: Vs. The Sinister Six, Peter marries MJ, Harry is Peter's best man, happy ending
Sinister Six is lead by Doc Ock and made up of himself, Vulture, Mysterio, Electro, Kraven, and Sandman.
