As far as one's past goes, a truly unique villain wouldn't be one with power, how many of those are currently running about. However, one that had a romantic or even peaceful past before hand and was simply walked out on or became twisted and evil and followed a path that their love could not follow or simply respect.

Another unique or even interesting type would be one that could write Ten Ways to Beat the Good Guy. By this I mean that they literally executed ten ways to beat the good guy.

Who noticed that Doctor Doom is all of the above, plus diplomatic immunity.
