The end of Superman : The Movie, with Christopher Reeve as Superman turning back The Earth to save Lois Lane. You can't beat that cinematic moment in comic movie adaptations, folks.
Reading the original Steranko and Kirby Nick Fury : Agent of S.H.I.E.L.D. stories; the original Lee and Kirby Fantastic Four stories; the original Kirby, Trimpe, etc. The Incredible Hulk stories; all give me this feeling. As well, the original Lee, Kirby and Romita Avengers stories; ALL original and early Iron Man, Thor and Dr. Strange stories; ANYTHING ELSE by and from Lee and Kirby, including the original X-Men; the work Kirby did at DC, including The Forever People; all Legion of Super-Heroes, Doom Patrol, Metal Men and Wildcats stories; War of The Worlds with Killraven; Marvels Kirby 2001:A Space Odyssey adaptation series and Mr. Machine / Machine Man series; the Deathlok : The Demolisher series; all the complete What If series; the original Punisher series; all series and stories featuring Captain America, Spiderman, Daredevil, Superman, Batman, Wonder Woman, Flash, Ms. Marvel, Black Canary, Iron Fist, Shang Chi, Captain Marvel, either one, and many, many other characters; all appearances by Modok, Galactus, A.I.M., Ego, The Mad Thinker and many, many other characters; that Marvel - DC amalgamated series with Super Soldier and Bruce Wayne : Agent of S.H.I.E.L.D.; all She-Hulk stories; and many, many other issues just send that chill through me that I am in a special place.

Sometimes it's the artwork, and sometimes it's the story, that appeals either to the writers or the artists, or to the readers. It's when they all work together, all the elements of the comic, for both the artists and the writers, and for the readers, that really makes the comic a work of brilliance.
Sometimes, the writing or the artwork alone can be so complete that it can overwhelmingly make the brilliance of an entire comic.
Of course, the readers ability to lose oneself in a comic can also make for the comics complete importance, even if the reader doesn't consider the artwork, or even the story, of the comic to be all that overwhelming.
Unbreakable is something I haven't seen but I'd really like to now. Hancock looks like another terrible decision for Will Smith. And, of course...
