Sinister is perhaps the greatest geneticist in the Marvel Universe. He is capable of cloning, creating superhuman abilities and enhancing or controlling mutant abilities. His existence is unknown to the general public and he does his research in secret laboratories across the globe. He has employed the henchmen groups the Marauders and Nasty Boys.
Origin

Nathaniel Essex lived during the 19th Century. It was the evil mutant, Apocalypse who turned Essex into Mr. Sinister. It was because of Gambit, and his ally Courier, that Sinister was able to gain the ability to shape-shift when they where stranded in this time period and required Sinister's assistance.
In the early 20th Century, Sinister was able to crack the genetic genome. During World War II, Sinster conducted an experiment in an attempt to clone the Sub-Mariner. The experiment went by the name of N2 and was defeated by Captain America. It was during this time that Sinister first worked with Scalphunter, a future Marauder.
Years later, Sinister used Scalphunter to steer a desperate Gambit towards him. In exchange for a surgery, Sinister had Gambit work for him. Gambit's final task was to assemble and lead the Morlocks into the Morlock Tunnels, initiating the Mutant Massacre.
Sinister also created a clone of Jean Grey. The clone went under the name of Madelyne Pryor. Sinister's plan was for Madelyne and Scott Summers (Cyclops) to have a child. It was his belief that the child of these two powerful mutants would become the ultimate mutant. Things between Scott and Madelyne went bad when Jean Grey returned from the "dead."
In a battle against Apocalypse, Scott's child, Baby Nathan, was infected with a techno-virus and the only way for him to survive was to go to the future. Baby Nathan returned as an adult known as Cable.
Later Sinister joined the reformed Weapon X program under the guise of Dr. Robert Windsor. He claimed to be helping mutants escape from Weapon X's mutant concentration camp but was, in reality, taking them to perform further experiments.
In Apocalypse's most recent bid for world domination, Gambit was chosen as the Horseman of Death. Following Apocalypse's defeat, Gambit and Sunfire fled to Japan to purge themselves of Apocalypse's influence. Sinister surprised them there and conscripted them into his service.
Sinister is currently working towards a solution to the Decimation of mutantkind and has allied himself with Exodus and the Acolytes in this endeavor.
