I think two villains is fine if you pair them right, they've already introduced Dr. Conners so they can use the Lizard. Another villain that would compliment the other would be Kraven the Hunter gets word of a giant lizard and comes to hunt it gets wrapped up in a three way fight as Spidey figures out the Lizards identity and has to save his friend from the hunter.
