There never is a formal answer. I thought this too before, because yeah Tobirama has red sage lines and white hair like Jiraiya. But Jiraiya never mentions it, and he talks about the 3rd Hokage all the time, so I feel like he would have mentioned being related to the 2nd Hokage. I want to it be true, but they also don't really share any jutsu other than shadow clones, which everyone knows. Jiraiya, Orochimaru, and Tsunade all have unknown last names, which I think is part of why they're so cool and unique- unlike every other character, they're their own person, separate of family, with their own unique abilities and powers. On their own, they rival clans from different hidden villages, which is insane. The Sannin are Legend.
