GLaDOS (Genetic Life-form and Disk Operating System) is the computer system that manages the Aperture Laboratory facility. GLaDOS released neurotoxin into the Enrichment Center and killed most of the staff. She goes on to be the main antagonist in Portal, Portal 2 and appears in the comic Portal 2: Lab Rat.
