around the biginning of The Other when Cap is teaching him to fight and he tries to catch a couple of bullets, ones goes through his hand :P big lols :P
in ultimate when he is confronted by the kingpin pulls a few pages out of his bag and starts reading prepared fat jokes i.e. your so fat you bleed cotton candy etc.. i almost wet myself
in new avengers issue 2 or 3 when they confronted electro and luke cage asked him how he used to fight him and spidey said he would web his hands up. luke told him to do it, but then electro passed out. so then luke told spidey to take it off, and spidey said it didnt come off, it would take about an hour. funnier with pics, but i cant do that where i am. also when they woke up in the savage land and spidey told everyone he didnt have underwear on. a lot of those early new avengers had good back and forths between luke and spidey. also the ultimate scene that hdorman1 was talking about with the prepared fat jokes.
I think some of my favorite moments with Spidey are in New Avengers, like brantumbo referenced. Some of the dialogue is fantastic and was truly some of the funniest stuff I've read.
On another note, I find Ben Grimm's dialogue some of the funniest, especially during Civil War(and oddly enough, Punisher-the war journal mini series). I don't know why but they are just too funny. From what I've read, dialogue contributes much more to humor than visuals.
theirs one where spidey is swinging through the city and is about to go through a window that is electralised and spidey says that he better not touch the frame or it would be curtains for him, it was a really bad joke but what made it funny was that he laughed at it and their was nobody else around
