What characters turned out to be the opposite sex?
From what I read, there was a mistake in the North American description of Birdo in the Mario Bros. 2 manual. The translation between the versions of the North American version suggest that Birdo is a boy that thinks he's a girl. That wasn't what Nintendo, intended for the character. Now depending on who works on games she's involved in, his\her gender varies. In Mario Tennis SHE is depicted as Yoshi's GIRLFRIEND. Apparently an issue of Nintendo Power describes Birdo as a SHE and in Japanese versions of some games she's featured in, her name is Catherine. All the Voice actors for the character have been female accept one. I think the verdict is still out though because there are still newer games that suggest her gender is male or a mystery.@Vance Astro: I read that Birdo was a transsexual.

Jerry, in the Tom and Jerry cartoon, will sometimes (a lot of the times) have amped up spikey eyebrows, plus the expression on his face when he's pleading for his life is often really feminine. Really can't blame me for doubting his gender.
Basically the only thing holding it together is his name, otherwise he can easily found his way into the other side, if the fans were convinced.

While Mike Bison is a very obvious parallel to Mike Tyson, I don't think he would have tried to sue them. The character is based on his likenesses, but he doesn't resemble Tyson and his backstory isn't reminiscent of Tyson's either.

That was strange I got a notification for this,it has nothing to do with me >_< weird
Though I don't know why OP is confused about Sesshomaru and Zero's gender. I think it was obvious that they were guys IMO.
