Probably too close to call, although in the race they did. Roadrunner was waaay ahead of Speedy, so much so that he was able to stop and wait for Wile E to fall and hit the ground and by near the end of the race they were only neck and neck.
You also then have the Roadrunner being able to move so fast he is just a blur, leaving consistent smoke trails and be able to make laps around Wile E with at one moment leaving him in the dust and then the next standing right behind him.
Speedy meanwhile, he was able to avoid bullets being shot at him, move faster than a rocket sled(although he admits that it was faster than him), outrun a car(it didn't seem to be really driving fast though, more casually). Speedy's speed though seems to vary at times.
So.....imo, too close however if I have to give it. Would give it to the Roadrunner due to consistency and more feats.
On a semi related note, the 3rd fastest toon I would have to say is Pepe Le Pew.
