The whole "Thanos x Death x Deadpool" triangle thing is like something straight out of an archetypal, cliche high school drama... consider this:
Thanos, the sheepish beta-male nerd who does everything for the girl of his dreams, Death, yet she rejects his advances
And Deadpool, the less-than-intelligent wild guy who treats her like a bitch and yet, she comes back for him...
Methinks that whoever wrote the whole Thanos-Death-Deadpool triangle is bitter about someone...

What's interesting to note is that Jim Starlin is one of Mistress Death's co-creators. She was designed to illustrate Thanos's dark desire for death. Her relationship with Deadpool is done far more as a joke than anything else, as she acts far different in his comics than she does in anything else.
Their three way relationship kinda sprung from that.
I think Deadpool is funny, but I personally prefer the Thanos and Death dynamic because it's so much darker and more fascinating. Death also acts like a cosmic entity (completely forget about that Thanos Rising trash) in her appearances with him, which I always feel is more fitting to her character.
Realistically, Thanos is the one who loves her more. He's killed billions/trillions for her and has taken over the cosmos several times in her name. He is also the official avatar of death. The interesting part, which was explored in the Thanos Deadpool comic surprisingly enough, is that Death purposely leads them on so that they kill more. What makes Thanos crazy is that he knows this and continues to go after her.
So to sum up, Thanos and Death=the original thing.
Deadpool and Death= a joke that's spread like wildfire.
