hardest bosses? let's see.. bat queen from DMC3, false savior from DMC4, nemesis RE2, U2/krauser RE4, Vaas FC3, dragon priests skyrim, covenent/flood/didact halo series, liquid snake MGS 3, i can go on an on..
Quickman from megaman 2. mysterious figure from khbbs(aka kingdom hearts three.) and Sephiroth from kingdom hearts 2 on the hardest difficulty.

I think there are harder ones in BL 2. Master Gee and Vermivorous to name a few. :)

I never played any DLC and me and my friends tried about a week on and off trying to get Vermi to spawn but the most we got too was the one before it and we never even got to that point again.

The BL 2 GOTY Edition will be coming out soon. More than 100 $ of content for far less. I would recommend that you give it a try at-least. Every DLC/extra content released till date. :)
The King Boo at the end of Luigi's Mansion, but I was pretty young. Took me hundreds of tries. OOH, or maybe Super Metal Sonic, or whatever that Metal Sonic is that races across the screen then turns into a blade. Yeah that. Again, I was pretty young, so games were still fairly challenging.
General Raam and Skorge on INSANE difficulty.
Cod 4 level where you have to wait for the helicopter on VETERAN (Though not exactly a Boss level).

Master Pabs on Monster Rancher 2. What made him so hard was the fact that you had to spend forever following a formula to raise a monster powerful enough to stand a chance, and if you messed up even one rotation then you basically wasted 12 hours of careful planning and had to restart from scratch :/

On top of my previous list, I'm adding the first Bowser fight from Mario on N64. If I played it now, it would be incredibly easy, but when I first played it, that was the first 3D game I played and the first game that had a joystick for your thumb to control your character. The controls were such a nightmare I must have died a hundred times before I figured out how to move and adjust the camera.
@whysosolid: I think this would fit better in Off-Topic since it doesn't really have anything to do with comics directly.
Anyway, I don't play many video games that much, but I'd have to say those Crab Droid things on Republic Commando take the cake for me. They where a nightmare to take down. Espesially when you have four or five SBDs pinning down the rest of your squad.

- Sephiroth from Kingdom Hearts 2 - KH 1 was worse IMO!!!
I never did beat Mike Tyson. I just couldn't get the timing right.
Sephiroth in KH1 was super tough, but I eventually beat him (after hours of trying). I could never beat Sephiroth in KH2 though, I probably wasn't a high enough level or something.
