In answer the thread title, Doom, Reed, Thanos, or the Doctor with an equal amount of prep should be a stalemate, or else barely a win for one side or the other. So any two of them together should be able to do it. Rassilon with prep removed magic from the universe/possibly entire multiverse, so he should be able to do it as well.
To your question in your post, it depends on what he does with the prep and if he knows who he is fighting. Prepping magic to fight Dr Insano is dumb, since Insano can shut down Classic Dr. Strange level magic. Something like, say, a mid to high level Supernatural Angel should be able to defeat him. Casually stop time, and then remove his lungs and heart. Unless he specifically prepped for that. Does he know what he is fighting or not?
TOAA, full powered Lucifer, full powered Michael, and quite a few others can beat him. Meta characters, who can leave their medium, can defeat him. Sure, he's amazing, but he's not omnipotent.
Also, since the OP doesn't state which version, shouldn't it be nu52 Constantine? Is he as good as memetic/old Constantine?

John almost never wins his fights with actual magic.
@jonny_anonymous: No, like, actually? So if I had a a sub machine and walked into a room he was in, somehow the bullets wouldn't hit him because he is more "ruthless"? Being ruthless doens't let you win unless you do something with that ruthlessness.
if no one lets him talk and make a deal and straight out kills him before he does anything, so where does prep get him, if u just mean in general if his prepared for the person his facing probably nil bcuz like Batman could beats lots of ppl if he has prep ahead of time, however if they can get past manipulation then alot of characters.

If john has prep he can easily outmanuever gilgamesh. he trickes Gabriel Hornblower who is close to his brother Michael and Lucifer in power and that is way more powerful than Gilgamesh and the whole nasuverse and are beyond dimensions, second only to God. he trick the first of the fallen and multiple hell lords many times, so gilgamesh unless he intends to straight up kill him John can win some round stop overwanking Gilgamesh its getiing old and annoying.
