The Winchesters go hunting in the Marvel Universe

In order to win the war against Raphael in heaven, Castiel sends Sam and Dean Winchester to the Marvel Universe in order to retrieve a cosmic cube. Unfortunately, a few creature of the night have been taken over by Raphael, aligned with Mephisto, to stop the Winchesters. The Winchesters must make it through all five rounds in order to retrieve the Cube.
All enemies are bloodlusted; Castiel has restored Sam's demon powers, Dean has been given the Muramasa blade(Cas has some connections!). Sam and Dean are fully healed after each battle and have knowledge of their enemies.(Apparently someone has been leaking superpower information on wikipedia)
Takes place in Central Park at Night.
Round 1: Vampires: Baron Blood and Bloodscream
Round 2: The Living Dead: Zombie(Garth) and Zombie Deadpool(full body, steel Katanas)
Round 3: The Pack: Werewolf by Night and Man-wolf
Round 4: Monster Mash: Frankencastle(no Bloodstone) and Nkantu the Living Mummy

Seeing that Sam and Dean have taken out their warriors. Raphael and Mephisto have deployed their trump card: The Ghost Riders! Castiel, sensing that the Winchesters are in trouble, sends some special help

Round 5: The Ghost Riders: Johnny Blaze and Danny Ketch
Sam and Dean are fully restored for round 5. Sam's demon powers are amplified ten fold and given an archangel blade(In this case, its similar to the Muramasa blade but can also channel angelic energy into blasts). Dean has his strength, speed, and awareness enhanced to Spiderman levels(current) and he is given the Colt with 12 shots.

Do the Winchesters survive their little hunting trip with the Cosmic Cube and return to their world, or do they fall to the might of Marvel's Mightiest Monsters?

I think you made round 1 far too difficult.  Vampires in the Supernatural universe are nothing compared to Blade and/or Morbius.  Either of them could take out the brothers.

If tyey pass rouns 1 i think they can make it to round 5. I wonder if the colt would work on the Ghost Riders

They don't make it past round one.

I've changed round one to Baron Blood and Bloodscream. Maybe it would be less difficult of a fight
