By your own scan, SoK killed the creator of his universe. The beings he killed were powerful, they just didn't do much since Preacher was not centered around feats like most Marvel/DC comic books.
What happens when SoK pulls the trigger on Lucifer (Going by the fact that the bullets never miss and always kill)?
The creator who was powerless without his chair. SoK admitted he wouldn't have been able to kill otherwise. He killed a god who was powerless, could die, was not omnipotent, couldn't even teleport to his chair.
On the other hand Lucifer is above death, he cannot be harmed by any physical means because he's just will. Always kill don't mean anything, what you might call kill didn't exist before Lucifer, the brothers were responsible for creating Death and they themselves cannot die.
