If the gladiator had any of the typical sets of weapons and armor used in the arena, the Spartan would slaughter him. Gladiators were given showy rather than practical weapons and limited armor so the audience would get a good show with lots of blood.
If they're equally armed this gets harder. Both have been well trained (the Spartan since he was 7) and brutally conditioned. A big difference here is that the Spartan is trained to fight in a formation (a phalanx) and the gladiator is used to one on one. The gladiator is fighting with weapons and armor not used in the arena so he's not as familiar with them as he could be, while the Spartan has lived by and with his weapons all of his life.
I can see this going either way pretty easily but I'm going with the Spartan because of his insane discipline and training.
