I wouldn't really say he is better than Luthor at prep in my opinion. However, what makes him deadly is his cunning and him being the best Con-artist (in my opinion) mixed with his prep. He can be pushed behind his limits and can be chased down by the devil like a lab rat and give him back his lung cancer and still emerge victorious. It's how he would deal with Luthor's prep and I'm pretty confident John would find a way to win.

John would win simply because he would be willing to sink to depraved levels to save himself that Lex Luthor would never even consider.

Lex. His intellect and the tech that he is capable of bringing to bear would be too much for Constantine. A mind-destroying psionic blast attuned only to Constantine's brain-waves that is constantly beamed over the entire planet via satellite hacking will do the trick. He could take John out in a myriad of other ways utilizing strategems both complex and simplistic.
