Well, you said that the team does not know where bullseye is, but spidey's sense or daredevil's radar, should be able to locate him.
Spider-man has shown to be able to dodge Lester's projectiles before. Now that he is using his abilities at their best, he shouldn't have trouble dodging his explosive projectiles. Spidey has dodged Gambit's cards before, so i think he should be able to dodge Lester.

And spidey's spider-sense has shown that it is able to detect his opponent's location.
The only power that i see that it will bring trouble to the team, it's Bullseye's healing factor. But in my opinion, spidey's punches combined with captain america's hits with the shield, should eventually bring Lester down.
But it will be a good fight.
