@Edamame:
If it's Smith from the first movie then it's anyone's match.

If it's Smith from the second or third movie he wins easily.

If it takes place in the real world then Smith is a floppy disk and gets snapped in half.

Elimination: Via "death"

Location of Battle: Basketball court

Blood lust: Off

I'll make it multiple scenarios here. Feel free to put what you think based on what you know.

To answer this question we must limit the set of scenarios and thereby establish a certain amount of rules that will be applied to both characters so as to determine the most likely outcome.

In essence, we have three scenarios:

Scenario A: Albert Wesker vs Agent Smith ~ First movie

Scenario B: Albert Wesker vs Smith (virus) ~ Second movie

Scenario C: Albert Wesker vs Smith (virus) ~ Third Movie

The rules are as follows:

  • Wesker has his Beretta 92F Custom "Samurai Edge" with 2 extra clips
  • Smith has his IMI Desert Eagle XIX with 2 extra clips
  • They are in character
  • Fight takes place inside the Matrix (otherwise the battle cannot be done) but Smith cannot use his assimilation or mind take-over of Albert Wesker in order for this fight to be 'fair'
  • Win is by death (as edamame states in original post)

With this in mind, because we know that they are in character, we know that Albert Wesker and Smith respectively, do not use their weapons in close-quarter combat. Wesker doesn't use his pistol unless its to combat B.O.Ws (Bio Organic Weapons - which are designed to 'soak' up bulletfire) or unless he absolutely needs to. Smith likewise chooses not to use his weapon in close-quarter combat as well. So, with this in mind, I judge it fair to say that Wesker and Smith's weapons will not be used further in this discussion when it comes to the final outcome, as both characters will choose not to use them in such a fight.

Now with that out of the way, let us examine their speed. Who is faster?

First, let us examine Agent Smith, and his instances of speed to try and determine just how fast he is

In the first Matrix movie during the helipad sequence Neo wields two Beretta 92FS pistols which have a muzzle velocity of 381 m/s. During that sequence, Neo unloads all his rounds at Agent Jones who swiftly dodges them all as evidenced in the below clip.

Loading Video...

Now, if we were to work with the assumption that all three agents (before the agents were upgraded in the 'The Matrix Reloaded') move at the same speed as each other, then it is safe to say Agent Smith is faster then 381 m/s. However, his speed is shown to be limited as he, Agent Jones, and Agent Brown were unable to dodge the bullets erupting from the GE M134 Minigun, which subsequently 'killed' them (albeit temporarily). Now the minigun in the movie (if we were to assume fires just as fast as its real life counterpart) has a muzzle velocity of 853 m/s, so it would be rather safe to say that Agent Smith cannot move faster then that.

Loading Video...

Now if we were to move towards his virus self in the second movie, we have quite little to go with as to how fast he is. However, what he do know is that since he still carries the weapon most commonly associated with Agents, the IMI Desert Eagle XIX, then it would be safe to assume that he still the same speed of when he was still an Agent. By the third movie, he moves just as fast as Neo so to answer how fast is Smith by the third movie, we must work out how fast Neo is.

Now, on the ground we can't really answer how fast Neo is because of lack of evidence, but we can work out (approximately) how fast Neo is in the air. And logically speaking, he should be just as fast, or faster on the ground, because there's less air resistance compared to when you fly above the ground.

We have two instances of Neo going all out in flight; to try and save his comrades/friends in the second movie. The first is when he races to save Morpheus and the Keymaker, and the second is when he rushes to save Trinity. Given the lack of distance numbers between Neo and Trinity during that sequence, we must use the instance of when he trues to save Morpheus and the Keymaker because of the greater regard for Neo's speed. When Neo calls Link about their location, he informs him that Morpheus and the Keymaker are "In the middle of the City, 500 miles due South." at 4:45 in the below video.

Loading Video...

Now it takes Neo about 13-14 minutes (depending on when you start counting in the movie), and assuming of course that every scene happens after the other, because if they didn't then Neo would be even faster then the calculations I give because it takes less time), so let us say 13 minutes and 30 seconds for rounding sake. Since Speed (m/s) = distance (m) over time (s) then we have something to work with. 500 miles is 804.672km which is 804,672m. It takes Neo 810 seconds (relatively) to reach Morpheus and the Keymaker. So: 804,672 divided by 810=993.422 (recurring). That means Neo can travel 993 m/s at his (presumably) top speed, assuming of course that he can react, fight and run at that speed on the ground when he's serious. It's a bit of a stretch, but it's something to work with.

Since Smith has been shown to fight Neo on equal footing in the third film, then it would be safe to conclude that their speed is roughly the same.

Loading Video...

So what does this mean in relation to Wesker? Is he as fast? How fast is Albert Wesker?

As shown in the below Resident Evil 5 Lost in Nightmares fight sequence against Chris and Jill, Wesker is easily able to react and dodge bullets while they're being fired at him.

Loading Video...

And also against Chris and Sheva in the Resident Evil 5 Rematch sequence

Loading Video...

Now Chris, Jill and (presumably) Sheva all wield M92F handguns as stated by the game. Now judging by their look, it would be safe to conclude that they are modified Beretta 92 pistols which fire with a muzzle velocity of 381 m/s.

In-game, Wesker is shown to be able to dodge all weaponry that the player fires at him (footage not necessary because if you've played the game, this is obvious). Now with that in mind, how fast are these weapons? Are there any that are faster then 1000 m/s? Let us name them all (in no particular order): Hydra shotgun and all other subsequent game shotguns (let us assume it fires slugs or bullets between 400 - 600 m/s), S&W M500 (632 m/s), S&W M29 (450 - 500 m/s), Skorpion VZ61 (320 m/s), SIG P226 (457 m/s), Heckler&Koch MP5 (425 m/s), SIG 556 (850 - 940 m/s), RPG 7 (115 m/s), H&W P8 (500 m/s), Grenade Launcher (70 - 80 m/s), H&K PSG1 (800 - 900 m/s), Px4 Hangun (360 m/s), Dragunov SVD (800 - 830 m/s). None are faster then 1000m/s but, if Wesker can dodge weapons such as the SIG 556 (without any visible effort) which can reach speeds of 940 m/s, then it would be safe to say that Wesker's top speed does exceed 940 m/s and perhaps 1000 m/s as well.

After all, in Resident Evil/Biohazard Code Veronica, he (presumably) dodged explosions which destroyed the Umbrella Antarctic facility, and point-blank range explosives which destroyed a house, as evidenced below in the comic: Biohazard Umbrella Chronicles: Prelude to the fall.

In any case, the only time where Albert Wesker was outmatched in speed (and strength), was against Alexia in Code Veronica. Does this mean that Wesker will also get manhandled by Smith?

Loading Video...

Section of Speed conclusion:Agent Smith from the first film and virus Smith from the second film can travel faster then 381 m/s but cannot travel faster then 853 m/s so their top speed is presumably in between these two numbers (381 - 853 m/s). This speed estimate will be used for Scenarios A and B. Smith was able to fight and match Neo in the third movie, so the speed estimate for Neo can be used for Smith in Scenario C.

In regards to Scenario A and B, Wesker is definitively faster then Agent and Virus Smith as proven by his dodging of powerful weapons such as the SIG 556 (940 m/s) which is conclusively faster then the GE M134 Minigun (853 m/s).

But in regards to Scenario C, due to their approximate closeness in speed, and no conclusive way of proving which is faster, the only way this can be settled is by their respective durability and subsequent strength. Because of their prowess in martial arts, it's safe to say that it will take more then martial skill to settle this, hence why it will be decided upon these two attributes. It's also worth noting that even if Smith were to use Flight as he did against Neo, he only seems to use it to give him more momentum behind his attacks, something Wesker is know to do as well (via running and jumping). So this fight will be decided upon their durability and strength.

So how durable is Agent Smith, Virus Smith, and full power virus Smith (first, second and third movie respectively)?

Now as Morpheus states in the first film, the Agents are bound by the rules of the system and because they inhabit the bodies of human beings, they cannot do anything beyond that which is humanly capable (i.e. they can't withstand lethal blows/blunt force trauma that the human body can't normally withstand). Because of this, Agent Smith cannot withstand things such as bullet-fire or blows that would severely damage the human body, such as broken bones (e.g. when Neo twisted his hand when he awoke as the One).

Loading Video...

What is different about Virus Smiths however, is that they all seemed to have a passive tolerance/immunity to damage as shown during the burly brawl sequence where Neo slammed a pole laden with solid concrete into a Smith clone's chest, and that same clone is shown slowly getting back up. In the same sequence he slammed the pole into another Smith clone, which sent it flying, and as with the other clones, it seemingly failed to damage it. Does this mean that they are immune?

Loading Video...

Do not be deceived, because upon closer observation, however, it would be safe to conclude that even the virus Smiths cannot withstand a bullet to the head because like with the Agents, he inhabits human hosts. So what this means is that in essence, Virus Smith is just as durable as he was as an agent but he has a passive tolerance for pain, not an immunity (otherwise he would be invincible because he would be invulnerable, but as we know, viruses are not immune to counter-attack).

With regards to full power virus Smith however, is that this passive tolerance for pain (he is a commuter program after all) grants him a greater ability in combat because as the fight with Neo showed, he lasted longer then Neo despite the two having similar strength and speed. He was more durable as a result (although not invulnerable as he did bleed).

To work out how durable full power virus Smith is, is to work out how durable Neo is. How durable is Neo?

As evidenced by the sequence in the Chateau, he managed to block a decorative wall sword with his hand, something which the very same Merovingian henchmen later used to cut through the legs of a (presumably) stone wall statue in a missed blow.

Loading Video...

And also, he managed to endure multiple blows from the army of Smiths during the 'Burly Brawl' sequence, who (presumably) were just as strong as when they were Agents (able to punch through solid concrete), with the only indication of damage being done to Neo, being his grunts of minor pain (he's still human after all). Now if we were to use this as a starting point, we can deduce that Neo cannot withstand repeated trauma to the body (he has higher durability then the first film, because his digital self is 'free' so to speak, but there is a limit to how much he take as when he awoke from the Matrix, he was puffing and visibly exhausted from the fight), for extended periods of time.

Loading Video...

Because Smith isn't human, he has no feeling of pain (hence the pain tolerance) so the damage can be withstood more then Neo can but the damage will still be applied. Does this mean that Smith will outlast Albert Wesker?

How durable is Albert Wesker? Has he ever tired? Is Smith more durable?

Now, we know that Albert Wesker can withstand point-blank range shots to the head (without any damage beyond some pain) as evidenced by the Resident Evil 5 Cinematic fight sequence aboard the plane and in that same sequence, he fell several thousand feet (maybe a thousand metres) with the only sign of damage being the loss of his upper-body clothes. This is all despite the fact, his body was weakened after being poisoned/overdosed with his virus serum stabilizer. Not only that, but he was stabbed by a combat knife straight into his forearm, and it failed to penetrate.

Loading Video...

However, as the game takes note of, Wesker needs to inject himself every hour with one of these serums, otherwise his body will destabilize from the virus that grants him his superpowers. Since Smith does not have one of these serums that will overdose Wesker as Chris and Sheva achieved, then for Wesker's durabilty to (presumably) lower, he'll have to fight for more than an hour.

Moving on. His high durability and recovery is further demonstrated during Resident Evil Code Veronica's ending sequence. Chris Redfield swung a metal bar straight into Wesker's arm, and it bent around it. Wesker then took a dozen or so steel girders (each most likely weighing 400 pounds, or 182 kilograms, so 3600 pounds in total, or 1.6 tonnes) to the face and only suffered a temporary daze (as evidenced by his walking and tone). During that same scene, he took a gas explosion to his (again) face, and only suffered minor burns (which by his next appearance, had healed completely).

Loading Video...

Not to mention how in Resident Evil 5, he is clearly able to withstand RPGs (weapons designed to take out tanks, airplanes and bunkers) to his (again) face and again only suffers minor damage (a temporary daze) as evidenced by the final fight in the hanger in Resident Evil 5. It's also worth mentioning how in that same fight, whenever Chris Redfield (a man who is at peak human strength at this point) punches Wesker in the face it hurts him, and he has to shake his hand because it's so painful, and this is from a man who had no qualms with punching a boulder into a volcano and could punch zombie heads to bits. But does this Smith is less durable then Wesker? Or does this mean Wesker is less durable then Smith?

Section of Durability Conclusion:With regard to Scenario A and B, Albert Wesker is conclusively more durable then Agent Smith from the first film and by extension virus Smith because of Smith's inhabiting of human bodies. He is still bound by the limitation of human people

However, with regard to Scenario C, despite Smith and Neo shown to withstand tremendous forces from being thrown, kicked or punched into the ground or through walls, it is highly unlikely that even at that point in time, that they can withstand the same amount of damage that Wesker is shown to be able to withstand, such as RPGs or point-blank range bullets to the face/head. So with this in effect, it would be safe to assume that Albert Wesker is more durable then Smith in all three scenarios. The fight will now be decided upon who is stronger.

How strong is Smith?

As mentioned before, Agent Smith is known to be able to punch through solid concrete and presumably virus Smith is also capable of doing such a similar feat. But if we were to go by Agent Smith handling of Neo in the subway fight sequence then we can determine that is about as strong these two Smiths get because it is shown to be humanly possible of withstanding (if you have great willpower).

Loading Video...

But what about full power virus Smith? Because of his fight with Neo, and how it has been described (and known) as the two being similar in power and strength, we must again ask ourselves how strong is Neo (when he was the One) in order to use that to determine how strong Smith is by the third movie.

Now probably the best instance of his strength is during the 'burly brawl' sequence when he shakes off a small 'mountain' of bodies which would be able 25 bodies.

Loading Video...

Now Hugo Weaving is about 6 foot 2 and the average weight of men that size is about 190 pounds, that would equate to 86 kilograms. Since there were 25 bodies, then 86 x 25 gives us 2150 kilograms or 2.150 tonnes. Because Neo initially struggled at first, then that should be around his maximum strength. Because the two are similar then it would be safe to assume that is roughly around Smith's maximum strength, but it would be a bit of a stretch to conclude on that since full power virus Smith had gained control of the entire Matrix. Whether that increased his overall strength is unknown but since his feats of power during the final fight were similar to Neo, then it would be alright to say that it didn't.

Is Albert Wesker stronger then Smith?

Now as demonstrated in the above "Resident evil CODE: Veronica X - Chris vs. Wesker [legendado]" clip, Wesker is clearly capable of punching through solid concrete (much like Smith) and like Neo and Smith, send his opponents flying with his punches or kicks (although more so in Wesker's case). As evidenced by his abilities in the Umbrella Chronicles comics and game, his kicks are clearly capable of tearing through flesh and bone, even advanced BOWs which are meant to have augmented endurance (i.e. Their heads can withstand bulletfire something which we've concluded upon that Neo and Smith could not).

No Caption Provided

His punches are just as strong, as evidenced by his killing of Spencer and Resident Evil 5's deaths. His hands can easily tear through human bone and flesh (even if it's reinforced with body armour), shown in the below clip link and video (3:25).

https://www.youtube.com/watch?v=w7BwbqeuZ80 ~ Spencer's death.

Loading Video...

I think it's also worth mentioning about his first treatment of Chris in Resident Evil Code Veronica. The encounter left Chris gasping for breath from the sheer force that Wesker gave.

Loading Video...

Section of Strength Conclusion:With regard to Scenario A and B, Albert Wesker is conclusively stronger then Agent Smith from the first film and by extension virus Smith, because Albert Wesker is shown to deal the same amount of force that they can, and more. Not to mention how they are limited by what is humanly possible, whilst Wesker is not due to his superhuman serum.

With regard to Scenario C, because of conclusive evidence showing Neo's strength, we don't how strong Smith is, but from what has been presented and worked out, it would be safe to say that Wesker would be stronger then Smith because he has shown to be able to tear apart flesh and bone with his punches and kicks, something that Neo and Smith were not able to do in the Matrix (even if we took into account the Superman/Dragon Ball-esque visuals).

Final Conclusion:

Albert Wesker will win Scenario A and B because of the conclusive evidence that points to his victory against Agent and Virus Smith. He is faster, stronger and more durable.

Albert Wesker's will also win against Smith in Scenario C but not as swiftly (or as assuredly) as it would be for A and B because of Smith's high pain tolerance, and flight (momentum attacks). Because of Wesker's higher durability and greater strength then this fight swings towards his favour. It won't be easy as they are both skilled fighters, cunning, resourceful and highly intelligent. They never let their guard down and look awesome in their sunglasses but in final conclusion, the winner in this fight of "Albert Wesker vs Agent Smith" then the winner is:

let's do the maths shall we?

1) weaker vs Smith(virus)-Second movie:

80 members of Smith's Clone Army were a real threat to the most powerful being in the Matrix(Neo) I mean they make Neo to Run! so these clones are really dangerous together, also they have gun all of them!, remember Wesker can dodge bullets from one gun but what if hundreds of bullets from smiths get fired at his head? weaker were shot at his head by Alice so he can't dodge all of them. Besides Smith can copy himself! that's his most powerful ability. just one touch is enough to kill Weaker and turn him into another Clone!!

Smith will win

2)Weaker vs Smith(virus)-Third movie:

oh boy, this one is good!, remember when smith punched Neo? what happened to the Matrix? that's right!, the layers of the Matrix split from each other!!! BC Smith has taken over Oracle so now he set rules for the world!! weaker never could punch so hard to split matrix"s layers.

Smith will definitely Win

3)Weaker vs Smith(Agent)-First Movie Part 1

smith can move to another body if he dies so, wesker needs to wins 7 billions battle with smith respectively in order to survive, can he do that?!!

Smith 99% will win.

4)Weaker vs Smith(Agent)-First Movie Part 2

now, this one is tricky, what if there is no other being in the matrix except One smith and wesker and wesker isn't plugged into the Matrix?

they both can dodge bullets, wesker can survive better at explosions and...! smith can punch concrete wall but so can wesker(I suppose), smith is sufficiently immune to punches,(remember Neo when they were fighting!).

OK, to solve this Problem we need to take a look at Neo, I'm pretty sure you know Neo is the most powerful being in the Matrix, now do you remember Neo when in the beginning of the Second Movie he fought with three Agent? those agents were upgraded, so lets assume they got better 20%, shouldn't a punch of Neo kills them? no it doesn't bc agents are pretty strong! even none updated ones(at the end of the first movie Neo Punched Smith but he remained alive)

Morpheus himself said:"you can win a battle with agents Neo,BC their Strength and Power is limited by the laws of nature and the Matrix" thus only Neo can do better than them BC he is an anomaly for the Matrix. Wesker is no anomaly. by conclusion I should say:

I believe Smith 65% will win but to be more flexible I should say Smith 40% will win, bc of weskers survival adaptation, I mean you can shot at agents and one bullet is enough to kill them but many bullets is needed for Wesker

The shockwave punch alone is beyond anything Wesker had in the strength department.
