The ghost of the original Dr. Jekyll posseses Dr. Spektor to recreate the infamous Hyde formula. A modern descendant, Howard Jekyll, takes advantage of the situation and uses the formula to become the evil and powerful Mr. Hyde.
A nefarious Fu Manchu-style villain, Dr. Tong will stop at nothing in his demented schemes for world conquest, even endangering his own daughter, the beautiful Lu-Sai. The devil doctor Tong finally meets his demise via his own treachery in The Occult Files of Dr. Spektor #24, leaving Lu-Sai at last free to seek her own destiny.
The infamous vampire tries to create a vampire legion to take over Transylvania, and doubtless his ambitions would not have ended there. Dr. Spektor defeats him with the help of his beautiful secretary Lakota Rainflower and the "cured" vampire Baron Tibor. Oddly, though Dracula escapes unscathed at the conclusion of the story, he never returns for a rematch.
Spektor first tangles with Victor Frankenstein's infamous monster when the fiend tries to steal Lakota Rainflower's brain for use in creating a mate. Spektor encounters the creature two more times, once battling him in Spektor's werewolf form and again with help from the barbarian Durak.
Kareena, a witch who serves the Dark Gods, twice tries to destroy Dr. Spektor, once using illusions of Spektor's worst enemies and of Lakota Rainflower, and a second time using a mind-controlled Dr. Solar, Man of the Atom. Though on their third meeting, Kareena professed her love for the Doctor and indeed helped him to save the earth from extra-dimensional invasion, Spektor was still left wondering if their next meeting would be as lovers or deadly enemies.
"Laird Ravenscroft" is actually an ancient sea creature possessing the body of the unfortunate man for over a century. The real Laird Ravenscroft is trapped in the form of the Leviathan itself, until it finally regains enough of his consciousness to telepathically explain its predicament to Dr. Spektor. Spektor engages the bet in human form long enough for the man in the monsters body to seize and destroy him, finally ending its own life of torment as well.
Ostellon is the Master of the Living Bones, an ancient sorcerer and enemy of Tragg and Dagar, both ancestors of Dr. Spektor. Revived in the modern world, he makes a pact with the Dark Gods to destroy Spektor, but is defeated with the help of a mystic spike carrying the magic of the Warrior Gods.
Ra-Ka-Tep is an ancient Egyptian sorcerer, mummified and entombed with a magic mask to trap his soul in his mortal remains forever. Revived in modern times by a graverobber, Ra-Ka-Tep takes the man's body and resumes his career in sorcery. Taking on his mummy-body once more to prevent the excavation of his tomb, the sorcerer almost succeeds in stealing the body of Dr. Spektor, but finally becomes trapped in his mummy form. He encounters Spektor once more in this form (this time in New York) and a final time in the Afterworld, where Spektor has been trapped.
Though not a head-on adversary of Dr. Spektor, Valdemar Van Helsing has posed a threat to Spektor's friend, Baron Tibor, and later to the Doctor himself. Descended from the man who originally destroyed Dracula, this Van Helsing has none of his ancestor's noble traits, and is a sort of "supernatural bounty hunter," tracking down and destroying otherworldly menaces for a fee.
